Putin dismisses head of Russia's troubled Dagestan region

MOSCOW (Reuters) - President Vladimir Putin has dismissed the head of Russia's southern Dagestan province, the Kremlin said on Monday, signaling concern over mounting insurgency violence and political rivalries in the mainly Muslim region.

Putin appointed Magomedsalam Magomedov to a role in the presidential administration, the Kremlin statement said, removing him from the post of provincial head that he had held since being appointed in 2010.

Ramzan Abdulatipov, a ruling United Russian party deputy from the region in Russia's lower house of parliament, was named to replace him as acting head.

Throughout the 12 years since Putin rose to power and crushed a separatist revolt in Chechnya, Russia has battled a simmering insurgency across its mainly Muslim Caucasus mountain provinces.

Dagestan is now the epicenter of militant violence in region, where suicide bombings, sniper attacks on officials and shootouts at road checkpoints area near daily occurrence.

Patients' own skin cells are transformed into heart cells to create 'disease in a dish'

Jan. 27, 2013 ? Most patients with an inherited heart condition known as arrhythmogenic right ventricular dysplasia/cardiomyopathy (ARVD/C) don't know they have a problem until they're in their early 20s. The lack of symptoms at younger ages makes it very difficult for researchers to study how ARVD/C evolves or to develop treatments. A new stem cell-based technology created by 2012 Nobel Prize winner Shinya Yamanaka, M.D., Ph.D., helps solve this problem. With this technology, researchers can generate heart muscle cells from a patient's own skin cells. However, these newly made heart cells are mostly immature. That raises questions about whether or not they can be used to mimic a disease that occurs in adulthood.

In a paper published January 27 in Nature, researchers at Sanford-Burnham Medical Research Institute and Johns Hopkins University unveil the first maturation-based "disease in a dish" model for ARVD/C. The model was created using Yamanaka's technology and a new method to mimic maturity by making the cells' metabolism more like that in adult hearts. For that reason, this model is likely more relevant to human ARVD/C than other models and therefore better suited for studying the disease and testing new treatments.

"It's tough to demonstrate that a disease-in-a-dish model is clinically relevant for an adult-onset disease. But we made a key finding here -- we can recapitulate the defects in this disease only when we induce adult-like metabolism. This is an important breakthrough considering that ARVD/C symptoms usually don't arise until young adulthood. Yet the stem cells we're working with are embryonic in nature," said Huei-Sheng Vincent Chen, M.D., Ph.D., associate professor at Sanford-Burnham and senior author of the study.

To establish this model, Chen teamed up with expert ARVD/C cardiologists Daniel Judge, M.D., Joseph Marine, M.D., and Hugh Calkins, M.D., at Johns Hopkins University. Johns Hopkins is home to one of the largest ARVD/C patient registries in the world.

"There is currently no treatment to prevent progression of ARVD/C, a rare disorder that preferentially affects athletes. With this new model, we hope we are now on a path to develop better therapies for this life-threatening disease," said Judge, associate professor and medical director of the Center for Inherited Heart Disease at the Johns Hopkins University School of Medicine.

Disease in a dish

To recreate a person's own unique ARVD/C in the lab, the team first obtained skin samples from ARVD/C patients with certain mutations believed to be involved in the disease. Next they performed Yamanaka's technique: adding a few molecules that dial back the developmental clock on these adult skin cells, producing embryonic-like induced pluripotent stem cells (iPSCs). The researchers then coaxed the iPSCs into producing an unlimited supply of patient-specific heart muscle cells. These heart cells were largely embryonic in nature, but carried along the original patient's genetic mutations.

However, for nearly a year, no matter what they tried, the team couldn't get their ARVD/C heart muscle cells to show any signs of the disease. Without actual signs of adult-onset ARVD/C, these young, patient-specific heart muscle cells were no use for studying the disease or testing new therapeutic drugs.

Speeding up time

Eventually, the team experienced the big "aha!" moment they'd been looking for. They discovered that metabolic maturity is the key to inducing signs of ARVD/C, an adult disease, in their embryonic-like cells. Human fetal heart muscle cells use glucose (sugar) as their primary source of energy. In contrast, adult heart muscle cells prefer using fat for energy production. So Chen's team applied several cocktails to trigger this shift to adult metabolism in their model.

After more trial and error, they discovered that metabolic malfunction is at the core of ARVD/C disease. Moreover, Chen's team tracked down the final piece of puzzle to make patient-specific heart muscle cells behave like sick ARVD/C hearts: the abnormal over-activation of a protein called PPAR?. Scientists previously attributed ARVD/C to a problem in weakened connections between heart muscle cells, which occur only in half of the ARVD/C patients. With the newly established model, they not only replicated this adult-onset disease in a dish, but also presented new potential drug targets for treating ARVD/C.

What's next?

Chen's team was recently awarded a new grant from the California Institute for Regenerative Medicine to create additional iPSC-based ARVD/C models. With more ARVD/C models, they will determine whether or not all (or at least most) patients develop the disease via the same metabolic defects discovered in this current study.

Together with the Johns Hopkins team, Chen also hopes to conduct preclinical studies to find a new therapy for this deadly heart condition.

(AP) ? Paul Pierce hit a go-ahead jumper with 31 seconds left and the Boston Celtics beat the Miami Heat 100-98 in double overtime Sunday then learned that All-Star point guard Rajon Rondo will miss the rest of the season with a knee injury.

Rondo will have surgery for a torn anterior cruciate ligament in his right knee. He was hurt late in Boston's 123-112 loss, also in two overtimes, to the Atlanta Hawks on Friday night.

The postgame mood in the locker room "was good" before he told his players the extent of the injury, coach Doc Rivers said, "and then it went down."

This game was the first in Boston for Ray Allen since he left the Celtics after five seasons and signed as a free agent with Miami. He scored 21 points.

Kevin Garnett had 24 points and 11 rebounds, and Pierce added 17 points, 13 rebounds and 10 assists for the Celtics, who ended a six-game losing streak, their longest in six seasons. LeBron James had 34 points for the Heat, whose winning streak stopped at four.

"As much as I've been a rival with Boston over the years, I never want to see anyone go down," James said. "It's terrible, not only for their team but for the league."

Pierce's basket gave the Celtics a 99-98 lead. James had a chance to put the Heat ahead but missed a 12-foot jumper with 6.8 seconds to go from the left with defender Jeff Green jumping out at him. Pierce got the rebound and was fouled.

He sank the first shot. Then, as a fan shouted, "This one's for Rondo," he missed the second.

Miami had one last chance, but Shane Battier missed a long jumper at the buzzer.

The Heat also could have won in the first overtime, but Dwyane Wade, who had 17 points, missed a long jumper as the buzzer sounded. They had led 93-89 after consecutive baskets by James, but Garnett hit a layup with 1:45 remaining and a shot from the right baseline with 1:14 to go.

Boston could have avoided the first overtime when Pierce inbounded from behind his backboard with two seconds left to Jason Terry. But Terry's shot from the top of the key was short. The Heat had tied it on a 3-pointer by James with seven seconds remaining in regulation after Allen missed a 3-pointer from the left corner with 15 seconds to go.

Rondo arrived at TD Garden on Sunday for a pregame shootaround but was taken to a hospital after complaining of knee pain. Courtney Lee started in Rondo's place.

The initial report was a hyperextended knee, but the team announced during the game that tests showed the ACL tear. His teammates didn't find out until after the final buzzer.

Allen was part of the Big Three with Pierce and Garnett starting in 2007-08. In their first season together, Boston won its 17th NBA championship. He played against the Celtics once before this season, a 120-107 Heat win in Miami in the opener.

The crowd gave Allen a standing ovation when highlights of his career with the Celtics were shown on the video board above center court during a timeout with 5:33 left in the first quarter. At the Miami bench, he raised his left hand in recognition.

Allen entered the game about a minute later and was booed when he touched the ball. The boos came down again when he took two free throws ? missing the first, making the second ? three minutes into the second quarter.

Allen went back into the game with 30 seconds left in the first half with the score tied at 43 and made a layup with 16 seconds to go. But Pierce sank a driving layup with 4.3 seconds remaining to tie it 45-45 at halftime.

It was Miami's first game in Boston since it won Game 6 of the Eastern Conference finals behind James' 45 points.

Turkish PM criticizes pre-trial detentions of military officers

ANKARA (Reuters) - Turkish Prime Minister Tayyip Erdogan has criticized the lengthy pre-trial detention of hundreds of military officers, suggesting it was sapping army morale just as Ankara vows to keep up pressure on Kurdish militants.

During his 10 years in power, Erdogan, whose party has moderate Islamist roots, has brought to heel the once all-powerful armed forces, which see themselves as guardians of secularism and regularly intervened in politics and carried out coups in previous decades.

Hundreds of serving and retired officers, including 20 percent of military generals, have been jailed pending trial since 2005 on conspiracy charges and plotting to overthrow the government.

But as initial public support for the investigations dwindles, with critics and even sympathizers saying cases have spiraled out of control, Erdogan has distanced himself from the trials.

"There are now close to 400 retired and serving officers inside. The most serious are accused of forming organizations or belonging to one. If the provisions for these are certain, then finish the job," Erdogan said late on Friday.

"But if there isn't certainty, then the hundreds of officers should be treated accordingly. This disrupts the entire morale of the Turkish armed forces. How can these people then fight terror?" he said in an interview with Turkish television.

While Erdogan has received praise for bringing the military under civilian control, the years defendants are spending in prison without conviction has raised fears the trials are a political witch hunt aimed at silencing opposition.

The first large-scale convictions came last September after a 21-month trial when more than 300 officers were handed prison sentences for plotting to topple Erdogan's government almost a decade ago. Hundreds more are still in jail awaiting trial.

Around 100 journalists are also in prison, as well as thousands of activists, lawyers, politicians and others. Most are accused of plotting against the government or supporting outlawed Kurdish militants.

Parliament voted to abolish the special courts used in coup conspiracy cases last July after Erdogan criticized prosecutors for acting as if they were "a different power within the state".

But the end of the special courts, established by Erdogan's government in 2005, will not affect existing prosecutions of the hundreds of military officers already in jail.

MILITANTS

Erdogan's latest comments also come after one of the most violent summers in three decades, with security forces locked in almost daily battles with militants from the Kurdistan Workers Party (PKK) and at a time of heightened tension with Syria.

Turkey has been one of Syrian President Bashar al-Assad's fiercest critics and has seen violence from the war in its southern neighbor spill into its own territory. While Ankara does not want to get sucked further into the conflict, it has threatened cross-border military action if needed.

Turkey's conscript army is the second largest in NATO.

The PKK, designated a terrorist group by Ankara, the United States and the European Union, took up arms in 1984 and more than 40,000 people, mostly Kurds, have been killed since then.

The state-run Anatolian news agency reported on Saturday the military killed more than 1,500 "terrorists" inside and outside Turkey last year, citing the country's general staff. Turkish warplanes regularly launch air strikes on PKK targets in northern Iraq, where the guerrillas have bases.

Reuters could not independently verify the militant death tolls and Turkey's military rarely talks to the media.

Security forces, including army and police, have taken heavy casualties over the past year with PKK militants stepping up attacks on convoys and outposts.

Hopes of an end to the conflict grew, however, after the government acknowledged state intelligence officials were talking to jailed PKK leader Abdullah Ocalan.

While backing moves toward peace, Erdogan has vowed military operations will continue until the PKK disarms, a stance Kurdish politicians say undermines efforts to build trust.

(AP) ? Venezuelan authorities promised a high-level investigation Saturday into a deadly prison riot that reportedly left dozens of people dead when National Guard soldiers clashed with armed inmates.

Vice President Nicolas Maduro called the violence tragic while speaking on television early Saturday and said Prosecutor General Luisa Ortega Diaz and National Assembly President Diosdado Cabello would lead the investigation. He and other officials didn't give a death toll from the riot Friday at Uribana prison in the central Venezuelan city of Barquisimeto.

"The prisons have to be governed by law," Maduro said.

Humberto Prado, an activist who leads the watchdog group Venezuelan Prisons Observatory, said inmates' relatives and media accounts put the toll at 55 killed and 88 injured.

The Venezuelan newspaper Ultimas Noticias and the television channel Globovision reported more than 50 killed, both citing Ruy Medina, the director of Central Hospital in the city.

Relatives wept outside the prison during the violence, and cried at the morgue Saturday as they waited to identify bodies.

Penitentiary Service Minister Iris Varela said Friday that the riot broke out when groups of inmates attacked National Guard troops who were attempting to carry out an inspection.

Varela said the violence had affected a number of prisoners and officials, but said the authorities would hold off until control had been re-established at the prison to confirm the toll. She said the government decided to send troops to search the prison after receiving reports of clashes between groups of inmates during the past two days.

The death toll provided by Medina rose late Friday after he had initially reported four killed and dozens injured. Ultimas Noticias reported that the victims included a Protestant pastor and a member of the National Guard, as well as inmates.

Opposition leader Henrique Capriles condemned the government's handling of the country's overcrowded and violent prisons.

"Our country's prisons are an example of the incapacity of this government and its leaders. They never solved the problem," Capriles said on his Twitter account. "How many more deaths do there have to be in the prisons for the government to acknowledge its failure and make changes?"

The Venezuelan Prisons Observatory said in a statement that in 2007 the Costa Rica-based Inter-American Court of Human Rights had ordered the government to seize weapons that inmates had in their possession at Uribana prison and to take measures to avoid deaths in the facility. The group called for the government to release a list with the names of the dead and wounded in Friday's violence, as well as details about weapons seized in the search.

"No one doubts that inspections are necessary procedures to guarantee prison conditions in line with international standards, but they can't be carried out with the warlike attitude as (authorities) have done it," Prado told The Associated Press in a telephone interview. "It's clear that the inspection wasn't coordinated or put into practice as it should have been. It was evidently a disproportionate use of force."

Prado's group says Uribana prison was built to hold up to 850 inmates but currently has about 1,400.

It was the latest in a series of bloody riots in the country's severely overcrowded prisons where inmates often freely obtain weapons and drugs with the help of corrupt guards. Venezuela currently has 33 prisons built to hold about 12,000 inmates, but officials have said the prisons' population is about 47,000.

In April and May, a prison uprising in La Planta prison in Caracas blocked authorities from going inside for nearly three weeks. One prisoner was killed and five people were wounded, including two National Guard soldiers and three inmates.

Two months later, another riot broke out at a prison in Merida, and the Venezuelan Prisons Observatory reported 30 killed.

In August, 25 people were killed and 43 wounded when two groups of inmates fought a gunbattle inside Yare I prison south of Caracas.

Chavez's government has previous pledged improvements to the prison system, but opponents and activists say the government hasn't made progress.

Varela, the prisons minister, said news media including Globovision and a local newspaper had done reports on the inspections, which she said had in fact been a "trigger for the violence."

Prado denied that, saying: "The problem isn't the work of the media. The problem is that the government hasn't disarmed the prison population."

Wind, snow thwart search for plane in Antarctica

(AP) ? Hurricane-force winds and snow were preventing searchers Friday from reaching a plane believed to have crashed in an Antarctic mountain range while carrying three Canadians.

Its emergency locator started transmitting late Wednesday about 680 kilometers (420 miles) north of the South Pole, but the weather has prevented search planes overhead from seeing the presumed crash site itself.

Rescuers don't know if the men are alive. Their plane has survival gear including tents and food.

The locator stopped transmitting Thursday night and crews have been unable to establish radio contact. Rescuers say a break in the weather is forecast Saturday.

One man on the plane has been identified as Bob Heath from the Northwest Territories, an experienced pilot in both the Antarctic and Arctic. Rescuers say the other two men were also part of the flight crew and that no passengers were aboard.

The propeller-driven de Havilland Twin Otter, was flying from a U.S. station near the pole to an Italian research base in Terra Nova Bay. Rescuers believe it crashed in the Queen Alexandra mountain range at an elevation of about 3,900 meters (13,000 feet).

Winds of up to 90 knots (104 miles per hour) have been blowing Thursday and Friday.

Steve Rendle, a spokesman for New Zealand's Rescue Coordination Centre, said rescue planes circled the area on Thursday and Friday but have been unable to spot the downed plane due to poor visibility. He said the battery on the locator beacon may have run out but that rescuers have a good fix on its location.

He said that when the weather clears, crews hope to establish a forward base at the Beardmore Glacier about 50 kilometers (30 miles) from the assumed crash site. He said there's a rudimentary runway and a fuel depot at the glacier.

For now, two helicopters and a small plane remain at McMurdo Station, the main U.S. base about four hours' flight away. He said the elevation provides extra challenges for helicopter crews.

Heath's wife, Lucy Heath, told the Calgary Sun newspaper that airline officials had told her her husband's plane was down, and she said she was just waiting for more news: "I'm so worried."

Bob Heath wrote on networking site LinkedIn that he typically spends this time of year coaching and mentoring other pilots in polar regions.

The missing plane is owned and operated by Kenn Borek Air Ltd., a Calgary firm that charters aircraft to the U.S. Antarctic program. In a release, the National Science Foundation said the plane was flying in support of the Italian Antarctic program.

Authorities from New Zealand, Canada, the U.S. and Italy are working on the rescue operation.

Antarctica has no permanent residents, but several thousand people live there in the Southern Hemisphere summer as a number of countries send scientists and other staff to research stations. The U.S. runs the largest program, with about 850 staff at its McMurdo Station and another 200 at its Amundsen-Scott South Pole Station, where the Canadians' flight originated.

One Tough-Talking Nerd: Bobby Jindal Brands Himself as Republican Reformer

CHARLOTTE ? The speech Louisiana Gov. Bobby Jindal gave Thursday to a gathering of Republican party leaders, released in advance by his office, was organized by Roman numerals. I. America is not Washington. II. How we win the argument. III. How we win the election. IV. Conclusion. Jindal's delivery resembled that of a nervous student rushing through an oral recitation of a term paper. The audience was as distracted as a room of high school classmates.

Which raises the question: Can a fast-talking, brainy policy wonk be elected president? Because even though Jindal told reporters after the speech that ?any Republican who?s thinking of running for president needs to get his head examined,? it?s clear the governor has 2016 in his sights. He?s currently serving as chairman of the Republican Governors Association, a frequent launching pad to a national campaign, and on Thursday, Jindal pitched himself as the guy who can lead the GOP out of political exile.

?If this election taught us anything, it is that we will not win elections by simply pointing out the failures of the other side,? he said.? ?We have to recalibrate the compass of conservatism.?? In a pointed jab at his party?s Congressional wing, he scoffed at their focus on decimal points and deficit spending in Washington, though didn?t propose an alternative for how to balance the budget.

?We seem to have an obsession with government bookkeeping,? he said. ?This is a rigged game, and it is the wrong game for us to play?We as Republicans have to accept that government number crunching ? even conservative government number crunching ? is not the answer to our nation?s problems.?

Jindal is best known for his intellectual chops and zeal for public policy. The Ivy League graduate and Rhodes Scholar led Louisiana?s health care agency at 24 and became the youngest-ever president of the University of Louisiana System at 28. As governor he overhauled the public school system, and now he?s trying to eliminate the state income tax.

But unlike most of his potential rivals in 2016 ? Florida Sen. Marco Rubio, New Jersey Gov. Chris Christie, Rep. Paul Ryan of Wisconsin, Kentucky Sen. Rand Paul ? Jindal lacks that all-important quality on the national stage: charisma. His rushed speech Thursday featuring tirades against the liberal media and Washington served as a reminder that he has not lived down his widely ridiculed rebuttal to President Obama?s first State of the Union speech in 2009. And after Mitt Romney?s resounding failure to connect with voters on a gut level in 2012, the Republican Party might look for a stronger personality in 2016.

Jindal was most persuasive when he urged the party to reach out to a broader swath of voters. Hispanic, Asian-American and black voters overwhelmingly rejected Republican nominee Mitt Romney, who was viewed as out of touch with their concerns.

?We must reject the notion that demography is destiny, the pathetic and simplistic notion that skin pigmentation dictates voter behavior,? said Jindal, the son of Indian immigrants who went on to become governor of a conservative, Southern state.? ?The first step in getting the voters to like us is to demonstrate that we like them.?

For now, Republicans are taking him seriously.

?Republican governors are the bright spot in our party right now, and Gov. Jindal certainly ranks near the top of the list,? said Wayne King, vice chairman of the North Carolina Republican Party.

??Charisma comes in many different forms,? said Robin Hayes, chairman of the North Carolina Republican Party, who served with Jindal in Congress and called him ?one of the smartest men I?ve ever seen.? Jindal?s approach, Hayes added, ?is probably more difficult than what people are used to with sound bites, but given the chance to earn people?s respect, I don?t think it will take him long. I would define him as an accomplished leader, father, innovator and doer.?

Jindal is also a risk-taker. A staunch opponent of abortion, Jindal wrote a Wall Street Journal column last month calling for birth control to be sold over the counter. He made the novel argument that selling it over the counter would remove religious objections to requirements that employers or insurers to cover birth control, and it would preclude Democrats from using contraception as a wedge issue to get women votes.

Don?t mistake Jindal?s outreach as a signal he?s moving to the center on hot-button social issues. Based on his record in Louisiana, he?d be one of the leading social conservatives in the field. He signed a law requiring women seeking abortions to listen to the fetus heartbeat, campaigned against a state Supreme Court judge in Iowa who backed gay marriage, and most recently, panned President Obama?s gun control agenda. Still, he portrays himself as a pragmatist.

"We?ve got to stop being the stupid party,? he said. ?It's time for a new Republican party that talks like adults. It's time for us to articulate our plans and visions for America in real terms. It?s no secret we had a number of Republicans damage the brand this year with offensive and bizarre comments. I?m here to say, ?We?ve had enough of that.? ?

2014 Honda Accord Plug-In Hybrid

Subtle styling differences define the Honda Accord PHEV.

The Honda Accord Hybrid is back and this time it comes in both plug-in and standard hybrid configurations. The standard hybrid will be produced in the United States and go on sale this fall, while the PHEV hybrid is already available, a 2014 model that went on sale at select New York and California dealers on Jan. 15. This midsize, front-wheel-drive sedan offers segment-leading fuel efficiency, besting the Ford Fusion Energi, the Chevrolet Volt and Toyota Prius PHEV models.

Electric-Only Power

The newest Accord model retails for $39,870 and can travel 13 miles on electric-only power. Its EPA combined MPGe rating is 115 MPGe, while its fuel economy rating is 47 mpg around the city and 46 miles on the highway. A federal tax credit of up to $7,500 is also available.

The new model uses Honda Earth Dreams technology, a two motor system that works with a 2.0-liter four cylinder gas engine to power the sedan. A 124-kilowatt electric motor does double duty as a continuously variable automatic transmission and is assisted by a lithium-ion battery pack. Regenerative braking also contributes to the electrification process.

Drivers can move between an all-electric EV drive, a gasoline-electric hybrid drive and a direct energy drive with two additional modes available to serve their commuting needs.

Default EV Mode

The default start mode for the Accord PHEV is all-electric, switching automatically to gas/electric hybrid mode when the battery state of charge reaches a bottom threshold level. A center console button enables drivers to choose EV-only mode, ideal for jaunts around the neighborhood or an HV mode that allows for regular hybrid operation. With the latter, the battery?s charge level remains constant.

Honda says that the PHEV Accord?s battery can be full charged in less than three hours using a conventional 120-volt household outlet via the sedan?s equipped cord. Charging time comes in at less than one hour when a 240-volt Level-2 charger is used. A smartphone app enables owners to monitor the charging state remotely as well.

Accord PHEV shoppers will find a car that matches the Accord Touring trim level while offering its own styling cues, especially evident across the grille. The new model is available in three shades including burnished silver metallic, the latter unique to the PHEV.

Ohio Manufacturing Plant

With limited distribution and a small number of models to offer at first, the Honda Accord PHEV will serve as a halo product for its EV fleet. The real test for Honda comes when the Honda Accord Hybrid is released, a model that will be built at its Marysville, Ohio plant, joining the Insight and Civic Hybrid when it goes on sale this fall.

NRA criticizes Obama's reference to 'absolutism' - NewsOn6.com ...

RENO, Nev. (AP) - A top National Rifle Association official on Tuesday accused President Barack Obama of seeking to redefine the rights of gun owners, telling a hunting and wildlife conservation group that the president's use of the word "absolutism" in his inauguration speech was an attack on law-abiding citizens who own firearms.

Obama said in his speech Monday that Americans shouldn't "mistake absolutism for principle, or substitute spectacle for politics, or treat name-calling as reasoned debate."

The remark was interpreted by the NRA as a reference to the organization's steadfast opposition to any new gun regulations.

NRA executive vice president Wayne LaPierre told a Weatherby Foundation awards ceremony that the Second Amendment gives Americans the unfettered right to own a firearm, but the president wants to redefine that freedom.

"Absolutes do exist, words do have specific meaning in language and in law," he said.

The president wants Americans to believe that "putting the federal government in the middle of every gun transaction" will make them safer, LaPierre said. But the NRA believes people have the right to defend themselves and their families with semi-automatic firearms technology, he said.

"No government gave them to us and no government can take them away."

Obama last week unveiled a set of legislative proposals and executive actions on firearms that were formulated in the wake of last month's Connecticut school shooting that left 20 children and six adults dead.

The NRA responded to the proposals by posting a Web video that labeled Obama an "elitist hypocrite" for allowing his daughters to be protected by armed guards while not embracing a proposal - supported by the NRA - that would place armed guards at all schools. The organization has also planned an aggressive lobbying push to thwart new gun regulations and has been raising money in response to the outcry for new gun laws.

LaPierre's speech on Tuesday was billed as a response to Obama's inaugural address.

PASCAGOULA -- The Pascagoula boys soccer team got one step closer to a return trip to Clinton on Tuesday, routing Brookhaven 8-0 in the first round of the 5A playoffs at War Memorial Stadium.

Mitchell Cecatiello scored three goals, giving him 25 this season, as the Panthers won for the 13th straight time. Pascagoula will host West Jones in a second-round game on Saturday.

"We started putting balls in early and kept the ball on the (Brookhaven) side," said Pascagoula coach William Denton, whose team lost to Starkville in last year's state finals. "We maintained control of the game, and got some young guys in to get some experience. I'm real proud of the team. We played well enough to win and move on to the next round."

Pascagoula went on top 1-0 just seven minutes in when Tristan Garriga drilled the ball in from nearly 35 yards out. Landry Lofton scored less than three minutes later to make it 2-0.

Cecatiello's first goal in the 21st minute put the Panthers up 3-0. Aaron McMillan and Cecatiello both scored late in the half to make it 5-0 at the break.

Angel Rosas and Griffon Sublett added goals in the second half, as did Cecatiello on a penalty kick. Panthers goalkeeper Jackson Graham turned away all four shots by Brookhaven, which ends its season at 8-8-1.

Pascagoula has not lost since a 3-1 defeat to Oak Grove on Nov. 8, which dropped them to 1-2-1 on the year. The Panthers have outscored their opponents 72-3 during their winning streak, with 10 shutouts.

"We're just clicking," Cecatiello said. "Everybody's playing their position well and our leaders are stepping up into their roles. We're just playing really well."

West Jones beat West Harrison 2-0 on Tuesday. Game times for Saturday will be set later in the week.

NASA's Alien Planet Archive Now Open to the World

Scientists with NASA's planet-hunting Kepler spacecraft have revamped the mission's online archive of alien worlds, opening up the database for the entire world to see.

Researchers are now posting all exoplanet sightings by the Kepler observatory into a single, comprehensive website called the "NASA Exoplanet Archive." Instead of going through the long planet confirmation process before making data publicly available, since December of last year, scientists have started shoveling out all the data Kepler collects into a comprehensive list.

"When we make that list, right away it goes to the archive," Kepler mission team member Steve Howell told SPACE.com during the 221st American Astronomical Society meeting in Long Beach, Calif., this month. "So the day we know about the list, the archive knows about the list. And then everybody, including us, can work on that list. But that list is dynamic so if we, or a community person, makes an observation and says, 'Hey, I looked at this planet candidate but it's really an eclipsing binary,' then that entry in the archive will be changed."

The archive has information about the size, orbital period and other metrics of any possible planet discovered and investigated by Kepler.[Gallery: A World of Kepler Planets]

"It's all in real time," Howell said. "The sausage-making process is exposed."

Before the new archive was debuted, astronomers were already doing creative work with the data. One created a visually stunning video of every known Kepler planet candidate ? 2,299 unconfirmed exoplanets at the time ? orbiting one central point.

The Kepler team?s new "open" attitude toward data release is giving everybody, not just members of the scientific community, a chance to do some hands-on scientific research by building their own experiments, Howell said.

A group of high school students has already taken data from groups of planets observed by Kepler and mapped them against a map of known stars looking for a pattern. Howell doesn?t think they'll see much, but he's glad that they have the opportunity to get creative.

"The entire world can help us with this Kepler data," Howell said. "I don't see any downside."

Planet Hunters, a collective of amateur astronomers, recently found 42 new alien planets using Kepler data that was publicly available prior to the launch of the new archive system.

It would be difficult for Kepler scientists to get their jobs done without the help of amateur astronomers around the globe, mission researchers said. By making the exoplanet archive more accessible, it could mean that more planets are found and confirmed in shorter amounts of time than ever before.

More people sifting through the wealth of data collected by Kepler means that scientists have a better chance of finding even more planets circling distant stars, Howell added.

Since its launch in 2009, NASA's Kepler spacecraft has flagged more than 2,300 objects as possible alien worlds. Although only about 100 have been confirmed, scientists expect that at least 80 percent could be certifiable exoplanets.

Currently, Kepler has entered a "safe mode" after researchers spotted an issue with one of the spacecraft's three reaction wheels that are responsible for orienting the telescope. The malfunction has stalled science observations, but researchers hope that the issue will be resolved by Sunday (Jan. 27).

Atari US files for Ch. 11 to separate from parent

NEW YORK (AP) ? Video game maker Atari's U.S. operations have filed for Chapter 11 bankruptcy protection in an effort to separate from their French parent company, which is filing a similar motion separately in France.

In a statement, Atari says the move is necessary to secure investments it needs to grow in mobile and downloadable video games.

Atari's U.S. operations have shifted to focus on digital games and licensing, including developing mobile games, and have become a growth engine for its owner. France's Infogrames Entertainment first took a stake in Atari in 2000. It acquired the remaining stake in 2008 and changed its name to Atari S.A.

But the U.S. operations have been better performing than the rest of the company. In fiscal 2012 digital and licensing revenue both grew significantly and contributed 70 percent of revenue, while sales in bricks-and-mortar stores declined.

In December, Atari S.A. said a credit agreement it entered into with investor BlueBay ? its main shareholder and only lender ? would lapse at the end of the year and the company was seeking other ways to raise money. It added that it expects to report a "significant loss" for fiscal 2012.

On Monday, Atari S.A. said it and its European operations would file related bankruptcy procedures in France at the same time as the U.S. bankruptcy filing.

CEO Jim Wilson said the moves were the "best decision to protect the company and its shareholders." The auction process in U.S. bankruptcy proceedings will "maximize the proceeds" going to shareholders, he added.

Creditors include accounting firm Deloitte & Touche, and retail stores Kmart and Wal-Mart Stores, although none are owed more than $250,000. Blue Bay is not listed as one of the U.S. operations' creditors.

Atari, which turned 40 last year, was a videogame pioneer with games like "Pong" and "Centipede," but has changed ownership several times amid financial problems. In its filing with the U.S. Bankruptcy Court in the Southern District of New York, Atari said it had $1 million to $10 million in assets and $10 million to $50 million in debt. It is seeking approval for $5.25 million in debtor-in-possession financing from investment firm Tenor Capital Management.

Atari said it expects to sell its assets or confirm a restructuring plan within the next three to six months.

Atari S.A., which trades on the Euronext Paris market of NYSE Euronext, has requested trading of its shares be suspended.

Developing microbial cell factories by employing synthetic small regulatory RNAs

Jan. 20, 2013 ? Biotechnologists have been working hard to address the climate change and limited fossil resource issues through the development of sustainable processes for the production of chemicals, fuels and materials from renewable non-food biomass. One promising sustainable technology is the use of microbial cell factories for the efficient production of desired chemicals and materials.

When microorganisms are isolated from nature, the performance in producing our desired product is rather poor. Metabolic engineering is performed to improve the metabolic and cellular characteristics to achieve enhanced production of desired product at high yield and productivity. Since the performance of microbial cell factory is very important in lowering the overall production cost of the bioprocess, many different strategies and tools have been developed for the metabolic engineering of microorganisms.

One of the big challenges in metabolic engineering is to find the best platform organism and to find those genes to be engineered so as to maximize the production efficiency of the desired chemical. Even Escherichia coli, the most widely utilized simple microorganism, has thousands of genes, the expression of which is highly regulated and interconnected to finely control cellular and metabolic activities. Thus, the complexity of cellular genetic interactions is beyond our intuition and thus it is very difficult to find effective target genes to engineer. Together with gene amplification strategy, gene knockout strategy has been an essential tool in metabolic engineering to redirect the pathway fluxes toward our desired product formation.

However, experiment to engineer many genes can be rather difficult due to the time and effort required; for example, gene deletion experiment can take a few weeks depending on the microorganisms. Furthermore, as certain genes are essential or play important roles for the survival of a microorganism, gene knockout experiments cannot be performed. Even worse, there are many different microbial strains one can employ. There are more than 50 different E. coli strains that metabolic engineer can consider to use. Since gene knockout experiment is hard-coded (that is, one should repeat the gene knockout experiments for each strain), the result cannot be easily transferred from one strain to another.

A paper published in Nature Biotechnology online January 20 addresses this issue and suggests a new strategy for identifying gene targets to be knocked out or knocked down through the use of synthetic small RNA. A Korean research team led by Distinguished Professor Sang Yup Lee at the Department of Chemical and Biomolecular Engineering, Korea Advanced Institute of Science and Technology (KAIST), reported that synthetic small RNA can be employed for finely controlling the expression levels of multiple genes at the translation level. Already well-known for their systems metabolic engineering strategies, Professor Lee's team added one more strategy to efficiently develop microbial cell factories for the production of chemicals and materials.

Gene expression works like this: the hard-coded blueprint (DNA) is transcribed into messenger RNA (mRNA), and the coding information in mRNA is read to produce protein by ribosomes. Conventional genetic engineering approaches have often targeted modification of the blueprint itself (DNA) to alter organism's physiological characteristics. Again, engineering the blueprint itself takes much time and effort, and in addition, the results obtained cannot be transferred to another organism without repeating the whole set of experiments. This is why Professor Lee and his colleagues aimed at controlling the gene expression level at the translation stage through the use of synthetic small RNA. They created novel RNAs that can regulate the translation of multiple messenger RNAs (mRNA), and consequently varying the expression levels of multiple genes at the same time. Briefly, synthetic regulatory RNAs interrupt gene expression process from DNA to protein by destroying the messenger RNAs to different yet controllable extents. The advantages of taking this strategy of employing synthetic small regulatory RNAs include simple, easy and high-throughput identification of gene knockout or knockdown targets, fine control of gene expression levels, transferability to many different host strains, and possibility of identifying those gene targets that are essential.

As proof-of-concept demonstration of the usefulness of this strategy, Professor Lee and his colleagues applied it to develop engineered E. coli strains capable of producing an aromatic amino acid tyrosine, which is used for stress symptom relief, food supplements, and precursor for many drugs. They examined a large number of genes in multiple E. coli strains, and developed a highly efficient tyrosine producer. Also, they were able to show that this strategy can be employed to an already metabolically engineered E. coli strain for further improvement by demonstrating the development of highly efficient producer of cadaverine, an important platform chemical for nylon in the chemical industry.

This new strategy, being simple yet very powerful for systems metabolic engineering, is thus expected to facilitate the efficient development of microbial cell factories capable of producing chemicals, fuels and materials from renewable biomass.

How to Start Over (Hint: It's Never Too Late)I got an email from a reader last week with the subject line "How to Start Over." In it, the reader shared his personal story: being about 10 years into his career, balancing life with kids, a family, and a big corporate job, all while trying to establish his own digital platform and business?and feeling like it's time to break free and change things, but feeling overwhelmed by the idea of "starting over."

He brought up a point that I've been confronted with many times. I wanted to share my response to his email, with one overwhelming preface: You're never "stuck."

As much as it may feel like you can't get out of your current situation, as much weight as you put on the importance of maintaining your nine to five, as much fear as you have of disappointing your family?regardless of your current situation and regardless of your age, you're never ever stuck.

Here was my email response:

"?I think you bring up a point that folks are faced with time and time again. I always share the story of my dad and I playing golf last year. He's been an accountant for almost 40 years, and he's always been apathetic to his work and has always told me he's just "working to retire". When I asked him why he doesn't quit and do something else, his response was that he's been doing what he does for so long, and it just "is what it is," and that he doesn't know what else he would do.

Believe me, I can understand the sentiment, and it's hard to get into something new when you've been doing the same old thing for so long. But it was this conversation that inspired me and continues to motivate me to not waste a moment that I have to work on doing what I really WANT to be (and should be) doing. It's kept me humble and active in continuing to build my business and establish myself professionally.

The best thing I can say is that you're not stuck. You never are. It's not easy to break free and do your own thing, especially with the added responsibility of supporting a family. But if I, a 27 year old with not a heck of a lot of experience, can get fired and start a business at age 24, someone at age 35, or 50, or however old, with a lot more life and work experience, can certainly do it. It just takes creating a plan of action for yourself, and not being defined by your age or held back by your current situation.

Take things one step at a time. You don't have to quit TODAY and go "pursue your passion." Create a plan. Start working on your passions on the side (it sounds like you already are), and set yourself up to make an easier transition in the not-too-distant future. It also REALLY helps to set a deadline for yourself?a date to take the leap?and work toward that?"

We're afraid of disappointing our family. We tell ourselves that we're not good at anything else except what "we do." We're terrified of change. We convince ourselves it's "too late."

The only way you'll be disappointed is if you never try. The only way to know if you can't do something is if you give it an honest effort. It's only "too late" when you die.

The best thing you can do is to start. And it's never too late to get started.

More hostages reported dead in Algerian 'final assault'

Algeria's special forces launched a 'final assault' Saturday to end a four-day-old hostage crisis. The state news agency said 11 militants and seven more hostages were killed.

Algeria's?special forces stormed a natural gas complex in the middle of the Sahara desert on Saturday in a "final assault" aimed at ending a four-day-old hostage crisis, the state news agency reported. It said 11 militants and seven hostages were killed.

The report, quoting a security source, didn't say whether any hostages or militants remained alive, and it didn't give the nationalities of the dead. It said the army was forced to intervene after a fire broke out in the plant.

The siege at the Ain Amenas plant, jointly run by BP, Norway's Statoil, and?Algeria's?state-owned oil company, transfixed the world after radical Islamists stormed the complex, which contained hundreds of plant workers from all over the world.

Algeria's?response to the crisis was typical of the country's history in confronting terrorists ? military action over negotiation ? and caused an international outcry from countries worried about their citizens.

The latest deaths bring the official Algerian tally of dead to 19 hostages and 29 militants, although reports on the number of dead, injured, and freed have been contradictory throughout the crisis.

The militants attacked the plant Wednesday morning. They crept across the border from Libya, 60 miles (100 kilometers) away, and fell on a pair of buses taking foreign workers to the airport. The buses' military escort drove off the attackers in a blaze of gunfire that sent bullets zinging over the heads of crouching workers. A Briton and an Algerian ? probably a security guard ? were killed.

Frustrated, the militants turned to the vast gas complex, divided between the workers' living quarters and the refinery itself, and seized hostages, the Algerian government said. The gas flowing to the site was cut off.

On Thursday, Algerian helicopters opened fire on a convoy carrying both kidnappers and their hostages, resulting in many deaths, according to witnesses.

Final communication

In their final communications, the militants said they were holding seven hostages: three Belgian, two Americans, a Japanese, and a Briton. They had threatened to kill them if the Algerian army attacked.

Algerian authorities estimated that about 30 militants occupied the Ain Amenas site Wednesday and with 18 already reported dead, it appeared Saturday that the hostage crisis was finally over.

The standoff has put the spotlight on Al-Qaeda-linked groups that roam remote areas of the Sahara, threatening vital infrastructure and energy interests. The militants initially said their operation was intended to stop a French attack on Islamist militants in neighboring Mali.

The accounts of hostages who escaped the complex highlight the cavalier attitude toward their lives taken by both kidnappers and the military.

Helicopter gunships used

Ruben Andrada, 49, a Filipino civil engineer who works as one of the project management staff for the Japanese company JGC Corp., described how Algerian helicopter gunships had earlier opened fire on vehicles carrying hostages and the gunmen who used them as shields.

On Thursday, about 35 hostages guarded by 15 militants were loaded into seven SUVs in a convoy to move them from the housing complex to the refinery, Andrada said. The militants placed "an explosive cord" around their necks and were told it would detonate if they tried to run away, he said.

"When we left the compound, there was shooting all around," Andrada said, describing the helicopters' attack. "I closed my eyes. We were going around in the desert. To me, I left it all to fate."

Andrada said when the vehicle he was in overturned, some of the passengers were able to escape. He sustained cuts and bruises and was grazed by a bullet on his right elbow. He later saw the blasted remains of other vehicles, and the severed leg of one of the gunmen.

Sprawling facility

The site of the gas plant spreads out over several hectares (acres) and includes a housing complex and the processing site, about a mile apart, making it especially complicated for the Algerians to secure the site and even find where the hostages were being held.

Casualty figures varied widely. The Algerian government now says 19 hostages and 29 militants have died since Thursday. Before the final assault, the militants claimed through the Mauritanian website ANI that the helicopter attack alone killed 35 hostages and 15 militants.

One American, from Texas, is among the dead, and the militants offered to trade two other American hostages for two terrorists behind bars in the US, an offer firmly rejected by Washington. At least one Briton, a Frenchman and Algerians have also died in the standoff. Escaped Algerian workers describe seeing people of many nationalities, including Japanese, shot down.

French Defense Minister Jean-Yves Le Drian said Saturday that there are believed to be no more French hostages in the gas complex. He said the Frenchman killed, Yann Desjeux, was a former member of the French Special Forces and part of the security team. The remaining three French nationals who were at the plant are now free, the Foreign Ministry said.

Statoil CEO Helge Lund said Saturday that there were only six Norwegians unaccounted for, from the 17 at the plant at the time of the attack.

Militants planned for months

The attack by the Mali-based Masked Brigade had been in the works for two months, a member of the brigade told the online Mauritanian news outlet. He said militants targeted?Algeria?because they expected the country to support the international effort to root out extremists in neighboring Mali.

The kidnappers focused on the foreign workers, largely leaving alone the hundreds of Algerian workers who were briefly held hostage before being released or escaping.

Several of them arrived haggard-looking on a late-night flight into Algiers on Friday and described how the militants stormed the living quarters and immediately separated out the foreigners.

Mohamed, a 37-year-old nurse who like the others wouldn't allow his last names to be used for fear of trouble for himself or his family, said at least five people were shot to death, their bodies still in front of the infirmary when he left Thursday night.

Chabane, who worked in food services, said he bolted out the window and was hiding when he heard the militants speaking among themselves with Libyan, Egyptian, and Tunisian accents. At one point, he said, they caught a Briton.

"They threatened him until he called out in English to his friends, telling them, 'Come out, come out. They're not going to kill you. They're looking for the Americans,'" Chabane said.

"A few minutes later, they blew him away."
